Pairwise-end sequencing reads both ends of each DNA fragment rather than one, supplying more overlap information for reassembling larger genomes.

Knowing a fragment's start and end sequence constrains where it fits among its neighbors more tightly than knowing one end alone.

Watch for

Pairwise-end sequencing yields more information per fragment than single-end shotgun sequencing, at the cost of more sequencing reactions.
